A combination of these ideas.
Maybe make the player the head of a state park with a large "town" area and a huge wilderness to explore beyond the boundaries.
I'd like for furniture to have more functions, like computers emailing villagers and friends, phones to call, etc etc.
"Shiny" bugs and fish, among other variants, that go for higher prices.
Villagers have improved AI, and actually do things instead of just being mobile conversation starters. For instance, you could see them actually catching fish or bugs (opposed to them simply having a rod or net out in NL), you could see them playing minigames without your involvement, and so on.

More Animal Crossing in general would be nice. Yeah tons of new items, and different kinds of towns, and MC faces, and other options like that.
Dating, I know it'll probably never happen, but it'd be SO FUN! Walk around town on dates, have more uses for the fucking flowers, go to Brooster's for coffee with them, have Valentine's Day matter as a holiday, hugging, romantic moments, have them cry their eyes out begging you not to leave them if you either break up with them, or stop playing for a long time and come back. I don't even want anything lewd, it'd just be so fucking adorable.
More things to do with the villagers is always nice as well. More in depth interaction with the villagers and normal NPCs like Sable and Isabelle. I would so pat Isabelle on the head if I could.
It makes me really sad to think we're probably not getting any of the stuff we want if another Animal Crossing comes out. But I'd settle for another Animal Crossing coming out.

I want to see the game be more like the movie they made back when Wild World was the newest game. Huge sprawling town and tons of geography changes and more in-depth activities would be big for me.
