You don't need to play SS1, but they do reference events from the first game. If you don't think you'll play SS1 then I'd suggest you read the plot summary on Wikipedia or something.
You can carry weapons, ammo, hypos (health/mana potions), research ingredients, hacking tools, etc.
Make sure you save regularly, as I remember it, SS2 only autosaves when you change floors of the ship, which doesn't happen very often.
Also note that the default controls are from another time and will probably be very awkward for you if you aren't used to old-school shooters. You can change the controls easily, but I've seen people bitch about them before.

I don't recommend any mods for a first playthrough. If you're absolutely a tremendous fag, you can use SHTUP, which provides increased resolution textures and doesn't fuck up the game's atmosphere too much. Don't you ever let me catch you using Rebirth, though.

I don't like most of the graphics mods because I feel like none of them get the atmosphere right. One of the strongest points for me in SS2 is how well they nailed the spooky, isolated, and hostile atmosphere. I've tried a variety of the graphics mods but none of them do a good enough job of capturing the SS2 atmosphere for me and I think they weaken the overall experience.

SS2 really showed me how important sound is for an atmospheric video game. It's really one of those games you should be playing with headphones at night, to get the full effect of the game's sound and atmosphere. The graphics are important as well but they take a back seat to the audio in this game.
