While an appearance by one DK rep wouldn't strictly preclude another, it does eat into Dixie's odds significantly. After Dixie, K Rool is the (significantly less) likely choice. While not relevant in recent games, Kremlings have a major presence in Smash. Cranky and Funky have had appearances in TF and MKWii respectively, and are not outside the realm of possibility.

Maybe you haven't played Xenoblade, but Japan did--and loved it, ranking it their 5th favorite Nintendo game of all time. A lack of presence in the US didn't stop Marth and Roy from making Melee, and it wouldn't keep Shulk out of Sm4sh. Shulk will likely appear again in the upcoming X for Wii U, and has excellent moveset potential. We've even seen multiple retweets from Xenoblade developers promoting Smash news.

Rhythm Girl is the easy second choice--she's on the cover of every game, and Sakurai seems to be making an effort to include more female characters.

Sakurai has said that he wants to give another retro character a makeover (like Ice Climbers and Pit) and with Takamaru's deconfirmation I think that it'll be Sheriff.
Sheriff was the first character designed by Miyamoto as well as one of the first arcade games Nintendo released. Since the character has never really been fleshed out, there would actually be a lot of creative freedom in reimagining the character.
Plus, the character would be completely unique among the rest of the roster and surprise everyone.

K. Rool is way more likely than you think.
