The opening is something like a warm and fuzzy afternoon and you see random slice of life moments, and in the end you see 2 kid's legs going upstairs and opening a door of a bedroom
then the ending music is the day fadding up, and ands putting figures in a box one by one.
The plot of the film follows the warrior Mario, the actor Kirby, and the soldier Link, all three guided by the god Palutena, on their journey to combat the evil sorcerer Bowser and his forces in a tournament to save Earth.
It'll be similar to one of those american reality shows.

MC is chasing a great evil force which is manipulating all the villians in a convoluted plan to erase existence, or something. Master Hand rips the characters and settings from different Nintendo universes and creates a new world with all of them.
They are to take part in a fighting tournament for his amusement, and the winner will be granted one wish and will be returned to their world, while the rest will stay and fight until the end of time.
After a while, many of the characters decide to join forces to overthrow Master Hand and use his Final Destination stage to return everyone to their homes.
Some, mainly the bad guys, oppose this because they want to dominate their worlds or something evil.
In the end the good guys are escaping Master Hand and his world on Final Destination thinking that they made it, when Crazy Hand catches them.
They fight him and lose, and they are badly injured, and the Final Destination stage gets sucked back to Master Hand's world.
Since they left, Master Hand had been torturing the baddies that remained, so they decide to agree to a truce with the good guys and destroy the hands and use the wish-orb to just wish they were all back home, good and bad alike.
After they win the bad guys double-cross the good guys and fight them for the orb and lose.
The good guys wish to return home and the bad guys are stuck in another dimension to fight over it until the end of time.

Characters are brainwashed from the beginning and after they're defeated they join whatever party beat them. The parties adventure through various Nintendo/ Sega/ Namco worlds, defeating the protagonist or antagonist of that world, until they get all of the Smashers together for a final battle against Tabuu or something. Guest appearances by assist trophies and collectible trophies, helping out per episode.

I can see two ways it would happen.
One is a slice of life anime where they all live together in some giant house and them getting into shenanigans, every episode would focus on 4-7 playable characters, and some times a special guest star, which would mostly be the ATs. Master Hand would be the landlord and force them into shenanigans. The Characters would also have personalities that are different from their normal selves, for example Captain Falcon would be shy to women, Link and Ganon trying to outdo each other to get Zelda's attention, Pit being a wet blanket.
The other would just be SSE, Series starts with mario on his quest to stop taboo or whoever, then on his way there he recruits more and more people, eventually the group breaks up to do w/e, all untill the end where everyone comes together to stop the bigbad.

Other characters have relationships with these characters but are not necessarily in every episode. Mario is never shown but implied to be Ikes over-demanding boss. Peach is Daisy's over-achieving sister. etc

The cartoon is set to take place after the games. In Rise of Lyric (WiiU) Sonic is self-centered and cares nothing for the team.

As the games progress he will learn the importance of working as a team.
Have it be like Log Horizon, with all the Nintendo characters wake up and find themselves in some new world, with bits of their own worlds here and there (Peach's Castle is the main location and base of the heroes). The main group the plot focuses on is Mario Link Pikachu and Kirby, with the four of them traversing around Smashworld searching for others from their worlds, meeting friends and foes from other Nintendo games along the way. Other groups form up too, kinda like in Subspace Emissary.
The kicker is that none of them can talk. Mario does his Italian gibberish from the Mario & Luigi games, Link is silent aside from the occasional grunts in combat, Pikachu should be obvious, and Kirby goes around with his Poyos. Voice-acting is very limited and most communication is done via gestures. Only a few characters (Fire Emblem folks, for example) speak fluently.
Plus since this is a focus on all of Nintendo, franchises that don't have characters playable can get representation too (Advance Wars, Golden Sun, Panel de Pon, Wonderful 101, etc).
The Finale isthe entire series was the machinations of two children, Sakurai and Miyamoto, playing with their trophies. The kids put down the toys at the end of the final fight and leave to go play video games. A zoom-in on Mario at the end shows that sparkle from the Melee opening

Things started getting weird around season 5, when there was an evil Zelda clone named Assassin J, who worked with dark Link, Majora came back disguised as a secretary named Madora, Link had to take care of a spoiled rich girl named Marshmallow, and there was an episode with a Christopher Walken parody talking about how he ended up in Link's world after being abducted by aliens, and wants Link to find his version of the triforce called the Unobtainium.

Looking at this thread, it's either SoL bullshit or some SSE-style story. Sad part is, that's likely all you can do without making it drag on excessively or having it become boring.
Master Hand, using a dimensional void pulls out Nintendo characters from every known corner. They're put into a large maze filled with traps, turns, etc. It's half the size of Earth with unique locales. Each character is on his own, but can team up if need be. Some characters settle in the areas they wind up in, others want out. Characters aren't just limited to playables, you get the villains(K. Rool, Black Shadow), Secondary and Tertiary characters, etc. Basically the trophy selection. Master mixes it up with the 3rd parties(only the ones confirmed or in past games)for extra kicks. Everyone speaks English(No poyo bullshit from Kirby or grunts from the Kongs)for the sake of consistency, even the Pokemon.
Whomever makes it to Master Hand then fights him and Crazy and then by winning you can send everyone back home or change the maze entirely from scratch. Basically Deadman Wonderland meets Survival meets Smash Bros.
