You'll miss some content, including two escape rooms, but a two-route playthrough isn't too bad if you just want a quick refresher. If you want to go through the two extra rooms, you could include the sub end.
If you really want to see everything again, you'll need to do the bad ends, since there are some exclusive conversations that only show up when you're not on the right route, but that's not a big deal if you're replaying.
