1. The Empire was NOT AWARE of the exact weakness in the Death Star the rebels were targeting. Remember, the aide informs Tarken they MIGHT have found "a weakness".
2. The exhaust port in question on the Death Star was not the only exhaust port. Seriously, how could anyone possibly think it was? A tiny 2m "exhaust port" servicing a battlestation the size of a moon? The reason it was so critical to the rebels' plan was due a flaw in the internal design of the station that allowed a blast in that port to set off a chain reaction. Note that the shot did not go "all the way to the center", the exhaust port was not a straight tube right down the middle of the ship-- that was only a diagram to explain things to the fighter pilots.
