Saint Seiya
Played in a campaign where everyone could draw on their birth sign. Signs were the deities, each with a portfolio, cult and powers granted. Powers started with a bonus to skills the signs were associated with. Getting more invested earned a bigger bonus when the sign was ascendent, but a penalty when its opposed sign was in power. At the highest level one could gain several minor powers when the sign was strong. Each was also tied to certain forms of magic, which could stop working entirely depending on the season.

If memory serves, in Tactics the Zodiac signs were Eldritch horrors that tried to invade Ivalice but got sealed into special stones by a living saint. The church rewrote history out of shame over them killing said living saint shortly after the last one was sealed, because the saint tried to tell people what happened (and I think for having a kid?) and wrote that the Zodiacs were mystical beings that aided the saint.
It was only after Ramza dealt with the horrors that they got re-sealed, and I think they were all banished back to their plane of existence after the last one tried to fuse with his sister (as their family has genetic ties to the saint... I think.)
It's been at least 10 years since I played Tactics, even if I've bought the re-release on PSP with the fixed translation.

On a more serious note, the zodiac signs play a somewhat important role in Ivalice, the setting for the Final Fantasy Tactics games.
I think one of them uses them as a mechanic where the relations between the zodiacs give you stat boni. Might also be one of the Tactics Ogre games, there's some overlap on the dev team.
Personally, I'd give each sign a domain, theme, etc. Every character has access to standard skills like swords or diplomacy, but their sign replaces or modifies those skills.
You could add interactions with stuff like other signs, the phases of the moon, and so on.
The rest would come from the worldbuilding, figuring out how societies react to those rules, what institutions form, and so on.

I tried something like this before, failed but that was more because I didn't know how to tell a story more than an info-dump.
I had the Zodiacs set up as usurper Gods which divvied up the land into twelve separate nations, with a couple of badlands thrown in there for entities stronger than the Gods (never got that far).
Their power was based upon the amount of believers they had, and they could bestow potential / power onto their believers in the form of Artifacts which varied in strength. These artifact bearers were basically the Gods' hands on the land, and over time their bodies would change to be more like their Gods'. Pisces brought on Naga, Capricorn brought on Goatmen, so on so forth. It also influenced the attitudes of the people, making some populations avaricious, hotblooded, or some obsessed with pursuing knowledge (some were assigned because it fit more with where the story needed to go).
I'd still like to do something with the setting but I've been chewing on something else for a while now. I had planned out a couple of the God battles in advanced, like fighting Libra inside their own plane of existence, fighting on top of a giant set of scales.

I'm in the process of trying to make something with this.
The solution I came-up with is to start with the zodiac and then branch-out into generally esotericism and new age mysticism.
I'm trying to go for a pro-rock psychedelia vibe to it with a lot of science-fantasy pulp elements. There's a lot of crystals, glowing shit and vaguely Greek architecture.
The signs themselves act as patron deities, codes of ethics and periods of time.
The signs are also divided into four groups which act as a character alignment system (for example, Gemini is associated with knowledge-focused characters while Virgo is for wealth-focused characters). This is based on the Hindu bhava system of astrology.

I don't like the divide by cities, especially not when each city is an entire civilisation unto themselves.
I mean yes, I understand that if we'll have people actively shun those of other zodiac signs, they'll inevitable want to separate as mach as possible. However I feel this level of separation wastes a lot of potential for plothooks.
Also if all the zodiacs have independent civilisations, that basically indicates that they all started worshipping stellar constellations with roughly the same associations and cult practices and each know how to put people of another zodiac sign into the context that they dislike them - But otherwise their cultures are absolutely independent?
I propose another approach:
Initially, there's this small city with a distinct local cult of worshipping stellar constellations, surrounding their zikkurat-temple. As time went on, so did the separation by zodiac, and people started to expand the city to allow for zodiac-exclusive residences, which eventually leads to people migrating to found smaller settlements that are entirely made up of single-zodiac population.
The capitol is still the old city, where the Zodiacs have clan feuds, but sometimes join forces in order to fight against another zodiac growing too influental, potentially through a baby-boom during that specific zodiac's birthtime.
The individual zodiac-clans still show the characteristics described in your posts, but they display them in their daily life inside their district. This fostered behaviour also leads to the zodiacs becoming something of a caste, where their birthsign not only determines their behaviour but also their profession.

Use a d12. Fix the stats and bonuses to work with it. Some rolls (perhaps magic?) would use the d12 to determine which zodiac sign was in favor at that moment, to determine the power or perhaps the effect. Your zodiac sign and the current zodiac sign could affect anything from stat bonuses/penalties to what might happen in the environment.
Zodiacs would have some interaction with each other, so that you know which zodiac signs are "allied" with yours and would give you a decent bonus. Perhaps spells (or even martial skills) would have zodiac alignments as well, where maybe a Taurus character would do well casting a Sagittarius spell, but have difficulty with an Aquarius spell.
Note that these ideas could work with Chinese zodiac just as well.
