New objective: Get a job that you actually like.

Why does everyone on this entire site act like there are only horrible jobs and everyone is a wageslave and everything is terrible in general, waaaah. Find something that interests you and expend some effort for once in your life. Weak willed pieces of shit.
>>
>>44823955
Wageslavery going well for you huh?

Revenge quest. Kick that one guy you know's ass. You know the one.

Can't do it with your fists? Get a flask of acid and call yourself an alchemist.

Next quest will be an escape quest, but first a little NPC romance...

It's more of an achievement system, really - education, wealth, social standing, stuff like that.
Presently grinding for better accommodation, need to get that upgrade by the end of summer so I can focus on education again. All while holding a job with a side of hobby craft (might drop that one for workout, not sure yet).

Mostly PC work, I don't do much lab work myself anymore. I personally find lab work fun though. Designing new primers, optimizing procedures etc.You know, actually accomplishing things, seeing actual results from work that you've done and not something that you do because someone told you to do it.
This is very satisfying to me.
Oh and the ~2-3 months a year dicking around in tropical regions to collect samples and conduct experiments are nice too.
I mostly just stare at a screen like this though. it's still fun, see if you can spot why I took this screenshot.

>>44824191
Soon anon, soon. Seriously though, with CRISPR becoming a thing and NGS soon to cost less than $100 you own domestic catgirl waifu might become both very real and very illegal in the forseeable future.

I took a year off and went to spend time with an uncle who was doing research in Vanuatu. By the time we moved to Fiji I was basically living as a beach bum. The shaman approached me and asked for help in reading English and I ended up going with him to help in that capacity after letting my uncle know. After that we traveled around the islands in a voyaging canoe as he helped local people with traditional duties such as funeral rites, while I helped him mostly with speaking and reading English but also however else I could. Supporting ourselves was easy enough off of basic survival skills which he was quite good at and taught me, but also the generosity of the people we helped.

Eventually it was more than just him and me on the voyaging canoe, and we ended up resembling and adventuring party for awhile I imagine.

Something that pays well and leaves you with enough free time for what you want. That's what I did, don't really care about my job but am happy.
>>
>>44824816
What's the problem Anon? Do you not take care of yourself?
>>
>>44823232
Ask everyone you interact to if they need help with something. Actively try to help them if they do, doing your best. Expect no reward but accept them when offered. Be it money, garbage or a meal. Interact with a lot more people. Learn everything you can on the way. Dont give up until the job's done.

That's how you quest in real life. Doing this I once gained a new fridge, and a later an angry handjob behind a gas station.
>>
>>44824463
Ok, the pig and the dolphin intrigue me. Elaborate please.

>>44824567
Not human no. It's stupidly specific.
Most amino acids in a protein are just "scaffold" basically, there are only a few actually important onces, the functional domain(s). What you're looking at seems to be the functional domain of that protein, in a short region the amino acids go hydrophobic > hydrophilic > hydrophobic (red > blue > red) and so on. That seems very non-random so it's probably important.
In my reference that pattern is strong but not perfect, there is one methionine in there that is not as hydrophobic as the others. In the species I'm assembling there is a single mutation that switches that methionine to an isoleucine, which is more hydrophobic and makes that pattern "better". This is pure conjecture but to me it looks like an advantageous mutation, which is fucking amazing considering how rare they are.

>>44824630
What do you do in your free time?

>>44824648

>>44824936
The pig belonged to the shaman, he had him before we met for the purpose of a feast though that ended up never being needed. The dolphin was wild and started following us and got pretty friendly with me, and I swam with it a few times.
>>
>>44825017
With all these disillusioned young muslims travelling there to fight in their holy war you have to wonder why there aren't also nutjob christians armed with whatever they can get their hands on moving out on a holy crusade.
No, that one guy with the sword doesn't count.

>>44825117
I don't actually know, I'm not a biochemist. I can only go by what I'm seeing, a very clear pattern of +-+- that is a little muddled in the reference at a little "better" in my assembly. I'd NEVER actually insist on that, this is just speculation.
Also I just looked up what that subunit actually does. Interestingly it forms the actual hole the protons go through. My guess is that this part forms part of the inner surface of that hole. That's only a guess though, I don't do protein folding.

I've had a few quests. One was to reclaim the family inheritance from my traitorous aunt by traveling over 3000km hitch-hiking across Australia in summer. I defeated her and won back the money my grandmother had saved her entire life so we could pay off the mortgage on her home. The snake retreated and I returned home a hero.

There was also the time I rode across California and Arizona to perform back alley surgery and dentistry on a mother and daughter on the verge of losing everything. I also fell in love on that journey, but it was not meant to be.

Get a job as a Natural Resource/Wildlife Bio major and spend your life travelling to different environs and parks.

Also take up Bow Hunting, and maybe visit an Exotics Ranch at least once in your life. It'll be super expensive, but the money goes to insuring that a species virtually extinct in its homeland doesn't go totally extinct on earth, and you'll be able to say you killed, ate, and stuffed your very own Kudu or Oryx without having to have ever done any unethical poaching.

Well, it's rare, but mitochondrial selective sweeps do happen. I know this because they screw up phylogenetic analyses.
>>
>>44834202
Well yeah, of course. But we always (read: if we have the time and money) use nuclear markers as well if we do phylogenetics.
I'm not one of those people that use CO1 barcoding data and thinks they can build trees from that.
I'm just building that mtDNA for fun. anyway.
>>
>>44823991
Wageslavery is a contradiction of terms. Only the dumbest specimen of socialist even actually uses the world or thinks it unironically.
>>
>>44834202

Your mum screws up phylogenetic analyses.

And actually if you see a string of alternating hydrophobic/non-hydrophobic what you're looking at is a section of protein that is largely structural - specifically forming a long straight section of the protein as the polar and non-polar residues try to maximise the distance between themselves by twisting and not allowing the protein to fold back upon itself.

The high proponderance of S, T and H residues furthermore suggests that we might be looking at part of a Beta Sheet, but the functionality of the site is doubtful when the low GC count of the gene sequence itself is taken into account.

Visit a monastery and gain a monk's blessing.

Plant a druid's grove of thirteen trees in a circle, make sure they survive to be taller than you are.

Move a boulder from one side of the country to the other in any way you can manage.

Court a young woman earnestly and truly, using Renaissance methods: writing an introductory letter to her father, then to her, etc.

Get someone to allow you to renovate a building. Do not require them to pay you anything.

Learn to build a weapon from scratch - any weapon.

There are no true limitations to real life questions except those you impose upon yourself.

I'll start.

Fuck off, you fucking braindead troglodyte.
>>
>>44835193
I'm being serious. I even worked at a temple for a time. You think all the monks do during meditation is nap? They do crazy backwards logic. They even pose those question to other monks who might not give them an answer that raises more question years later. The whole idea is that life is a big circle and they strive for the mind to do so as well.
Whats the sound of one hand clapping?
-3 years later-
But master...why was the hand clapping in the first place?
-wise nod-
-7 years later-
Why do we clap?

Enlightenment is the ultimate state of being fusing mental, spiritual, and physical being. Humans can't reach enlightenment because we aren't ultimate beings. We compound this with being enlightened at birth and letting life corrupt us with moralities that are swayed by masses and ideologies that can only be explained with words which lack the inherent ability to wholly explain things as they truly are. Seriously ask a monk and you'll get essentially the same answer it IS monk 101.

I'm with you. I teach high school physics, and it's the best fuckin thing. I would do it for free if I was otherwise financially taken care of. The job itself is fun and usually easy, and it makes me feel great about contributing to society in a meaningful way. I get summers and weekends off to travel with my family too. I actually met my wife with this job (another teacher), so our schedules always sync up.

Anons, there are such things as good jobs. You just gotta figure out what you want out of life and then figure out how to get payed for it.

>>

Apparently my family stopped some time during WWII and the Vietnam war before saying fuck it and left for the US.

>>44835109

The key word there is "mostly" - nuclear and cellular pores often use beta sheets within transmembrane channels and pores to act as gates or filters, other times they're used to form especially large pores. But more likely it's part of a spacing motif that seperates more functional aspects of the protein, or part of a section that connects and holdd in place other proteins as part of quarternary super-structures on either side of the membranes it passes through.

For someone whose country doesn't do student loans, how does it work? You get loan on the virtue that you'll use it to do college and pay it back after you get a job with that degree, but if you don't get work with a degree you don't have to pay it back? Some sort of loophole? How comes they give you another loan if you haven't paid back the first? Won't they repossess your property to cover the loan if you ever get any?
