Stop, step back. You're getting ahead of yourself.

First, what do you want to make? A setting, a rule set, both?

Second, Why do you want to make it? Is this just a general 'That sounds fun', or do you have a specific idea, vision or concept you want to make work?

Third, why would a game you made (or adapted from a base system) be interesting? What about it would make it different from other games you've played, why would people want to play it?

Ask yourself those three questions, in great detail. I don't even care if you answer them now, just think about it. They're absolutely fundamental to any bit of RPG design.

OP here...this would be more of a creative exercise for me, something where I'm more interested in setting and character than I am with creating systems. I know that I could use any one of a thousand existing systems, but this is kind of an excuse to flex some world-building muscle.

Why do you even care about the system if you want to do worldbuilding? Just do all the worldbuilding you like, if you want to play games in it when you're done just snag a generic system then, don't think about making the world fit the rules in advance, that always leads to shit worldbuilding.

Okay, you need to clarify some terms here.
You want to make a SETTING, not a ROLEPLAYING GAME, and you're wondering which game you should use for your setting. This is kind of a weird thing to ask, as most games aren't explicitly tied to their setting, and even then, they can usually be written in a way that separates them. The core of a game system is usually just a mechanical way to resolve problems.

So just pick whatever game you like, and spread your setting all over it.

You should check out Fate. It's a generic, CC system with genuinely flexible rules (as opposed to GURPS' incredibly inelegant you-can-do-anything-because-there-are-rules-for-everything approach), centered around the idea of creating your own character traits and abilities. It's notoriously love it or hate it and can be an absolute trainwreck in the hands of the wrong group, but it's also free if you get the ebook, so there's no reason not to check it out.

ORE is the most flexible and fastest classic structure RPG. It is a bit on the gamist side, but easily incorporates obsessive detail rules if you are so inclined.

Fate is the meta machine where rules don't apply to the physics of the game world, but to the story.

Both run fast, come with a large community and countless mods to steal from, and have a free core game to download.

If you want to keep it really simple and micromanage the details as GM, look at BRP. All it does is resolve skills in d%, including combat.

These are all generic. But for simple ideas it is usually better to custom tailor crunch that does just one thing, but that very well. Look at Don't Rest Your Head for truly well made mechanics with a great inner dynamic that directly translates into story.
