Quality post.

I skimmed the wiki; is there anything unique about the setting? Seems like your average sci-fi dystopia with a spunky attractive young female protagonist.

Its similar to Bastion. It has a good sound track.
If you like Bastion you will probably like Transistor.
The final boss is an amazing character.

You can't really run a game set in Cloudbank with out just using the conflict with the Process and the Camerata again. Cloudbank's whole thing is that most of its citizens are happy and all the real issues stem from things the Camerata caused.

Most of those issues that they had were caused by the Camerata or the Process.

Platt was looking for her friend and was killed by the Process.

Moyle was killed while joyriding in the Process District.

Yondale, the Inspector, and most of the others ended up as functions because of the Camerata.

Not true at all. The whole point of the Transistor was that it collected individuals who were too exceptional for society and felt stifled by it. There was the not!football player who was such a strong hitter he ended up hamstringing the very sport he loved with rules made specifically to stop him from injuring more people, or the public works administrator whose humanitarian projects were constantly rejected by society

The Camerata are kind of a metaphor for fascism.

The core members were frustrated that everyone being allowed to vote on everything resulted in everything being bland and uninspired. If some of your friends hate vegetables and some hate meat, the group will always settle for plain cheese pizza every time they order; in the same way, the polls of cloudbank kept resulting in the same sunny skies, the same bridges, the same parks, ect...

The Camerata's solution was to gather special individuals from around the city, lock their souls in the transistor, and make IT run the city, believing that those "best and brightest" would know what was best for the people better than the people did.

Getting back to the OPs topic-
There would be several difficulties running a game in transistors setting, including:

1. Figuring out "What IS transistor's setting?" the VG deliberately leaves it ambiguous weather the people are some kind of TRON-style sentient programs or people in a virtual world. We also never really learn what the transistor is or how it works or where it came from. When your players ask these question, what will you say?

2. No visuals- that art nouveau aesthetic is a really important selling point of the game, and unless you are a really good artist with markers on a vinyl mat, that will be hard to bring across to players.

3. How many players do you have? Will each character get their own transistor? Will there be characters inside transistors? What would a character inside a transistor even do besides commentate?

The important elements in Transistor are rather conducive to a social/intrigue campaign.

1. Cloudbank is literally ran by committee.

Your players are movers and shakers: They can call influence through their audience, fans and disciples. This can be reflected through an system like Noblis or a more abstract work.

2. Cloudbank is dying of stagnation.

Parks give way to boardwalks give way to bridges give way to parks.

Something needs to change in the city. As it is, there is no progress nor development: Why be different when you can have the same good thing? Viva la difference... or don't, and the Camerata will ensure your influence will shape the city... once and only once.

The process is a natural antagonist, but also a powerful tool guided by the transistor: Why bother archiving society when you could raise the blade high above the streets and, once and for all, declare "Parks", and never again see the gaudy vaudeville or the grotesque architecture?

We're a civilized bunch, so I'm sure no one would hurt anyone for even ultimate control... They'd just convince their opponents the city is not for them; There's a place for them in the country after all!

The setting is great for a RPG, but not that much is specified, so if your GM is up to the task, go for it.
It would only really work for social campaigns (unless you introduce hostile elements), there are no weapons anywhere, no war, little strife except the riots that Red caused with her songs.
Set your players up as a Camerata like group: movers and shaker trying to achieve something specific.
Forget the Transistor itself (unless you want them to "discover" it), you can introduce any kind of mystical digital-ish artifice and pseudo magic as plot points and to spice thing up.
Remember that only the (previous and current) wielders of the Transistor can even see the Process without special (and perhaps even nonexistent) tools.

Just because it happened does not mean it's supported.
It was a bittersweet ending. Everyone's dead, but Red now has the power to rebuild Cloudbank and maybe even make new people to inhabit it if she tried. Instead she makes the selfish choice and commits sudoku to be with her BF, with little guarantee it'll work.
You're supposed to make your own conclusions and opinions about things, anon, instead of accepting that everything shown is good or supposedly good.

I'll admit, when I first unlocked the Agency tests, I thought it was referring to The Agency as a noun, rather than as an adjective. This led me to believe that there was some sort of Post-Cloudbank Agency that kept safe the infinite number of other virtual worlds, each agent armed with a Transistor. Pretty far off from the game but the mind does strange things with a misinterpretation.

I didn't interpret it as a selfish choice, perhaps because I don't believe that Red had the power to make new people. A new city, yes, but not new people. The most I believe she could have done was recreate a new Cloudbank and then live there alone forever except for the disembodied voice of her dead lover.

It's like a paranoia where Friend Computer is composed of the vote of every citizen on every matter, including the colour of the sky, and where instead of commie mutant traitors being reviled, it's the latest unfashionable thing.

Cloudbank has this weird thing going on where the city is in stasis despite everything being in flux. Visionaries of all stripes do their best to make it something beautiful, but not always successfully. There's a clear understanding, reading about Cloudbank, that it isn't all or even mostlybad; the impermanence leads to a lot of beauty. The probably is that a lot of the time things that should be permanent aren't, while things that could stay healthy in a state of constant flux are the things that don't get changed, like the constantly-blue sky.

Of course, not changing the sky is innocuous, and it kind of becomes part of the identity of the city. "The sky looks blue because we want it to," as the saying goes. It could be any other colour, but we choose this one as an act of affirmation. Still, sometimes rain might be nice.
