I have before.
I used Traveller but each character had a fourth layer of Health representing Kinetic Barriers their suits or Biotics generated.
Biotics I mostly based on Psionics since they're really just a slightly-more-plausible stand-in for space magic anyway.

there's a pretty well fleshed out fanmade Fate hack for it. It's too big for me to post the pdf here, but i'm pretty sure there's only one mass effect Fate system. It's given a lot of satisfying crunch to an otherwise kind of light system, while maintaining Fate's emphasis on narrative. I ran it for about a year and never once regretted it. Only trouble is that the pre-made enemies aren't quite properly balanced. It's generally better just to stat up your own after a while so your encounters aren't just piss-easy all the time.

I'd use quite a bit of the TL10 equipment from Ultratech, some of which would need trimming due to the universe's infinite ammo rules

Get very immersive, serious focus on player choices, with many callbacks and long-term consequences, have NPCs comment on things the players have done. Really weave a rich tapestry of the game world.
Then, right before the final fight with the BBEG, cancel all future sessions, and make the players pick one of three similar epilogues via e-mail.
If they complain, grudgingly add a fourth similar epilogue option.
