Well there are many ways it could be true.
Number 1, Twilight dies in a sacrifice.
Option 2, they all die at the same time in a sacrifice. They could also ascend into the afterlife instead of "dying" to tone it down.
Option 3 They give the other 5 immortality somehow.
Option 4, They all become alicorns.
As option 4 is incredibly unlikely, and I do think the staff would be thinking about a way to keep their statement true, Option 2 or 3 seem likely.

I think it should end with two of the Mane 6 reflecting on how their lives have changed over dinner, years later after the previous episode or the movie, showing us how the Mane 6 reached their goals and moved on, giving us all one last lesson on friendship when the rest of the Mane 6 come in for a surprise reunion, showing friendships last forever. The episode ends with them all in a hug, and the book closing and Celestia saying they all lived happily ever after.

The Finale for S4 would have been an incredible ending to the series IMO.
It had a great epic feel to it, gave a feeling of "completeness" to the six's paths as the EOH bearers with the throneroom appearing, and the song just gave a great wrap up to the series.
The S5 finale was shit for ending the series, and the S2 finale would have been terrible, but the S4 finale was GREAT for ending the series.

Also if there was to be a season to follow, it might have been better if the show focused on a different cast of characters, different location maybe. It didn't matter who as long as the spirit of the writing from season 1 would still be in place. It would give us a different perspective on the world of Equestria itself while simultaneously allowing for more merchandise and content. The writers also wouldn't be driven into a corner since they have a blank slate to work with.
