It's because the Princesses know that they're in a cartoon show, they know that no matter what happens, no matter how bad it gets, something will occur that will save the day. They have literally no worries because they know that no matter what, their side will win, and no one dies because it's a kid's show. This is why instead of trusting things to skilled and competent subjects and specialists, they trust someone who is untrained and unskilled other than being book smart.

My personal theory is that Celestia, Luna, and Cadence could've probably dealt with Tirek as a team with Twilight, but decided to let Twilight handle it on her own after she sang about wanting something to do and to be taken more seriously as a Princess. Celestia was probably just like, "Ok, then."

But why Twilight specifically? The reasoning was the fact Tirek didn't know about Twilight's Alicorn status, which is a bit of an oversight considering Discord knows all too well about Twilight and they left a big stained window dedicated to her in the same room they decided to stay in.
Celestia, Luna or Cadence could have taken the power, unless perhaps Twilight doesn't know the transfer power spell.
Even then, Discord being part of it is still sort of strange, since they know what he's capable of and surely the first thing he'd do is take away Twilight's elusive box linked to the tree of harmony

The princesses were caught off ward and it's implied they were heavily drugged (the same way Twilight got nearly captured), the vines also feed from magic, of which applejack has very little compared to her physical strength (even if we account part of that as magic-induced); if you take 5% of the blood from a lemming he's going to become a bit dizzy for a while but will live; if you take 95% of blood from a whale you're going to have a very dead whale.
The sisters are also much bigger than the ponies (square-cube law) and much slimmer too, they are also very well used to their magic and may or may not be very lightweight to sustain flight -even with their unusual wing-body size ratio-. So I wouldn't really count on them for feat of incredible physical strength either.
