Starlight mentioned that she "studied magic" so perhaps she is just part of a larger organization orchestrating this? If the writers aren't completely shit at continuity they probably remember stating MULTIPLE times that cutie mark magic is hard/dangerous. Since even Twilight seemed convinced (at first) that the staff was removing the cutie marks, it could be a rare spell. Also the fact that she got away at least implies it's possible for her to return.
Could be interesting is there is an actual season-wide "anti-friendship" league the Mane 6 has to combat.

I like it. Its like how Larson slowly introduced Starswirl the bearded. It builds up so much depth to the lore. First Twilight dressed as him in S2.04 then there was a library section named after him in S2.20 finally for him to be important to the plot when twilight solves his spell in S3.13 (all those eps written by Larson)
Mage Meadowbrook is like the next Starswirl I think. Maybe older, or just from a different place. All we know is Twilight described him as an "Eastern Unicorn" which makes sense since the S5 village was in the East of Equestria. First he (or maybe she) is first only mentioned in name, maybe later we will see him in a flashback or something.

This is where she finds solace, as a tinkerer with machines.
Anti-Pinkie: Pinkie's source of happiness is external, she finds happiness in making others happy.
Perhaps the anti-Pinkie searches inward for her source of happiness, but never truly finds it. Her search for passion becomes her passion, and as result her personality is philosophical and distant.

You could still make the list work. Twilight's passion before she met the others was learning about magic. The same is true of Sunset. But they had different approaches. Sunset was greedy and wanted all the knowledge and powerful magic to herself; she was all about power.
The difference between Twilight and Sunset, is that Sunset decided she didn't need friendship, and ended up being power-hungry and manipulative. Whereas Twilight, (apprehensive at first and initially dreading the prospect), embraced her friendship with the rest of the mane 6, and became the element of friendship/magic.

I think we're assuming we've met these characters before. It's most probable that they're new characters to sell toys.
I've been saying since S4 ended, that the only way to top Tirek in terms of threat level was to get a Legion of Doom going, using either old or new rivals of the Mane 6. The leader could be a G1 villain since they've been bringing them back recently.
Also the powers-that-be have pretty much confirmed Starswirl is showing up at some point with all the setup they did in the journal. It may not be this season, but at some point he is popping up, and that gives access to time-travel story shenanigans. You can do big stuff with that; they might even save it for the movie.
