It doesn't really matter with modern file systems. Single large files will be marginally faster.

Windows disables the write cache for removable media by default, which means small files are going to be very slow.
When you enable write caching, there is no difference to an internal hard drive however.

Linux enables write caching by default, so there's no difference to internal HDDs.
