The infinity gauntlet controls every aspect of the universe. Thanos is one of the few beings who actually know how to use all of the gems. With it he literally controls all of space time.
In the comics, Adam Warlock receives takes the gauntlet and lives through multiple timelines before he starts to go insane. A meeting is called between the great cosmic beings(living tribunal, chaos, order, etc..) they are the beings that control the universe, they force him to split up the gems because the gems combined are more powerful than any of them.
