The thing is, there are several ways to do this. In the comics, the Gauntlet gives him supreme power to manipulate the cosmos, but there are other forces who can do this too, so Thanos has to do battle with them as well. The remaining Avengers, Adam Warlock, Doctor Doom, and others who go up against Thanos only get any hits in at all because Thanos lowers his power a bit to show Death him beating them in "fair" combat, in an attempt to impress her suggested by Mephisto. Thanos is supreme in his abilities, but is still Thanos, with all his psychological failings, and that proves to be his undoing ultimately, with him merging his consciousness with the universe, but being unconsciously unwilling to remove his physical body from existence, so Nebula takes the Gauntlet off the hand of his physical form.

The only thing it CAN'T do in the comics, is to affect anything in a different universe. There is a gauntlet and stones in each universe, and each offers infinite power in that universe alone. If you try to use the stones to affect a different universe, it would have some effect but the stones would be destroyed as a result.
This rule is what stops villains from going around the multiverse collecting stones by the bucket load. Because there is an infinite amount of stones and gauntlets in the multiverse.

Yeah. Thanos is one of the race called Eternals, created by Jack Kirby.
A sub group of Eternals left Earth and settled on Titan.
Also, Drax was originally a human (a saxophone player named Arthur Douglas) killed by Thanos. The Eternal space god called Kronos took that human's pissed off soul and crafted a new Anti-Thanos body for him. Drax is more or less immortal and will always reincarnate, albeit in a slightly different form than his previous iteration.
Moon Dragon was Arthur Douglas' daughter, who was present when Thanos attacked and was taken by the Eternals and taught crazy mental skills by space-monks.
None of that can be true for the movie version though since Drax is apparently just some random alien.
