They both were scientists that caught on fire while working on a plant-effecting chemical and then fell in a swamp and mixed with the chemical and turned into a plant/swamp monster.
They debuted within like a couple months or so of each other so no one is sure who ripped off who.
The only real difference is that Swamp Thing has a human-like brain/thoughts and Man-Thing is a largely unthinking monster who's brain is all fucked up. Plus Man-Thing makes shit catch on fire if it's afraid of him.
Moore added some crap about how Swamp Thing was actually a plant elemental that just THOUGHT it was human, but that all got retconned away by New 52 and now it's back to the way I described about.

Im pretty sure that Alan Moore stuff is canon with Snyder's run. Alec meets the Swamp Thing before him and the whole point of the beginning was Alex coming to terms that he actually had to BE Swamp Thing this time.
I haven't read the new run though so unless it undoes that the Alan Moore stuff is still canon.

True, I dont think DC really cares. But going by the beginning of Snyder's run Alec was revived in Brightest Day and was being called by the Parliament to become Swamp thing for real. He had memories of the Swamp Thing that thought it was Alex Holland but eventually became a man inside the Swamp Thing someways in. Later on he becomes full Swamp Thing without himself at the center but still his proper consciousness and actually meets the original monster who thought it was Alec Holland. So unless the new run goes against that Alan Moore's idea is still canon, just in the past.
