Spongebob would be about the best DM you could expect from a character, and JC Denton's love of reading and philosophy would make him an ideal player.I have no clue what Goku would do though. I only chose him because he is the only anime character I have even the tiniest amount of knowledge about.

Pizzaz is a police chief and an easily pissed off one at that. The Boss is the leader of a gang that was able to gain so much power and popularity that they made themselves international celebrities and were eventually able to get themselves into the White House, with The Boss as Commander-In-Chief. So I would imagine there would be some conflict and tension between the two. Both of them would try to backstab or screw each others' characters over. Outside of the game the two would have loud arguments and throw insults at each other (the Boss would probably make fun of her name). Kintaro would attempt to mediate peace between the two.
At first, Kintaro won't be incredibly good at playing D&D. In fact he'll probably be pretty bad at it. However he would progressively better as he plays more, learning more about the game. Also, he would become attracted and get pervy over Pizzaz, as he does with every beautiful woman he meets. This of course, causes Pizzaz to get very *cue Venetian shades* mad at him. However she does eventually warm up to him because even though Kintaro can be a pervert, he still tries to be nice to and develop a genuine relationship with a woman he meets. As for how Kintaro is with The Boss, he doesn't really have any problems with him. Kintaro is friendly to The Boss and The Boss would be friendly to him.
Eventually though, Kintaro does something incredibly stupid, either ingame or outside of the game, which causes Pizzaz and The Boss to get super angry with him, causing him to have to leave. Later however they realize that Kintaro had done something that was able to help them out greatly. So Pizzaz (who has realized that she has fallen for Kintaro) and The Boss realize they were wrong to push Kintaro away and they go look for him. However Kintaro is already long gone from where they are, riding his bicycle looking for the next new experience.

Maka runs game but is really freeform about it because she's more interested in telling a story than the game mechanics. Hank insists on applying math to every situation which frustrates her to no end. Dante is just there for combat, eating pizza and making jokes in the downtime.

I assume the way the question is worded that the co character is GM, so we get a terribly convoluted plot that leaves all character development behind, but sure is one fell of a wild ride. Haruhi would create an impractical High Elf Sorcerer Princess with a LA of +7 from some obscure or homebrewed templates.
Shodan would desperately try to include some Omnificer cheese with liberal rule interpretations, forsaking Pun Pun as she'd be required to be a divine minion.
Double D's quaint and purposefully bland storyline of subduing inflation of grain prices in a Dwarf town turns into a bloody genocidal campaign across the multiverse from Shodan and Haruhi trying to out-god each other. Double D would sometimes comment on how their conquest effects gold prices, nervously trying to railroad them back onto his originally campaign idea.
