Leo, the Mothra in Rebirth of Mothra Trilogy, is referred to as a male and officially listed as a male in Toho's materials.
And how she reproduces is depending on the version, they all do it a different way. Mothra in Showa Era was implied to absorb enough radiation(Because everything is radiation then) to make it possible... because babies are kind of like growths, right? That's how that works?
Heisei Mothra is literally born of the Earth, same as Battra. She doesn't lay eggs herself, the Earth lays a moth egg. And a spikey, edgy moth egg. Because reasons.
Tokyo SOS is a mix, mystic, spontaneous pregnancy.
GMK mothra is a mystic guardian ala Heisei, she's born of the earth to defend it.
Leo is actually implied to be the last in a long line of mothras, and there used to be a fuckload of them. You get to see all the ancient mothras in ROM3, when they seal Leo in a cocoon to gestate over the course of millions of years and become Broken-Ass-Cheap-Fucker-Mothra. Leo probably had a dad.

Not Rebirth of Mothra version, they had more than the one back then, Leo is just the last one. There was a ton of these guys back in the day.
