No. Lemon works great for fats, Vanilla works great for fats, but they shouldn't be combined unless it's a rare exception like perhaps cheesecake.

Depends on the dish, amount of each you use. I imagine the lemon would be quite overpowering in most cases. For a cake maybe put vanilla in the cake and then make some icing with the lemon juice and grate the zest over the top?
Lemon's a great flavour on its own, vanilla and lemon brings to mind curdled milkshakes.
Just make a lemon drizzle cake.

Any citrus with vanilla is good.
Maybe except for grapefruit.
