I've thought about this for a long time, to the point people call me autistic, but honestly? I would put my money on Kenshiro. Jotaro has a definite chance though. Ken doesn't tend to activate his abilities without first gauging the strength of his opponent. Jotaro uses Time Stop when he needs it, just frequently enough that I could see him doing it before getting any hit in.
After heavy thought, I believe with the way fighting spirit/energy ("touki") works in Hokuto no Ken, it would basically make him able to punch stands if applied to the Jojo logic of fighting spirit = Stand power.
So for all those bitching, ORA vs ATA is definitely something that could believably happen.
If Ken survives the first hit or Jotaro doesn't do a killing blow with time stop when the fight starts, it's over.

1. Stands don't exist in Hokuto no Ken
2. Stands in Jojo are formed from "fighting spirit" basically
3. In Hokuto no Ken, fighting energy/touki manifests physically as auras and various attacks
It's all ultimately fanwank but it's certainly believable that suitably strong HnK characters could face off against the average power-type Stand in JJBA based on rules from both.
In the event that none of this would work, Kenshiro could activate Musou Tensei and be intangible to all but the most powerful demonic energy (whatever that might mean in Jojo terms; probably not Star Platinum at least) effectively making it a rock vs a hard place.

As awesome as the fights are and as much as I love Bruno and Mista, they really do not spend a lot of time on the characters at all.
At the very best you may get a page after a fight and maybe half a page before.
And it's not like SC where during the fights they add little flecks of character here and there, in VA all the fights are so intense that nobody does anything other than try to win and stay alive.

If you really look at it, towards the final fight Diego was basically made to develop Valentine, he's the front runner when it comes to facing off against D4C, establishing its strengths and weaknesses and nearly overpowering him to show just how weak he can truly be. After Diego vs. Valentine there's no way Johnny wouldn't have won against default D4C. Diego's cunning and strong will nearly overpowered him but ultimately Valentine's own strength came on top, someone like Johnny would put up an even bigger fight even without unlocking the infinite spin. Love Train came just in time to save Valentine's ass, but johnny still came on top.
This also makes it thematically fitting that AU Diego would be the force behind Valentine's last will, his presence alone reinforces Valentine's cunning and distrust for Johnny

Part 6's lack of character interaction is 100% because of the setting. You can't just have people in prison having a good old time chatting with each other when they're under constant surveillance and tight schedule. I've always really liked Diavolo for some reason, but I can clearly see why a lot of people are bored by him. I just like how completely insane he is, and how he manages to be a driving force in the story without sitting on his ass like DIO. That scene when he reveals himself to Polnareff, and by extension us, made everything 10x as tense for me. Even as Giorno miraculously heals the crew after every fight, Diavolo still manages to make them drop like flies whenever he shows up.

Well his problem (Dio and Kars have a similar btw) is that most of his development is offscreen. Dio has the luck that part 6 really fleshes things out and explain behavior in part 3, Kars has shit luck for being in a very short part and being overshadowed by his companions, but Diavolo only has half of already short part 5 to really flesh things out.
That and more than half of his development goes to Doppio instead.
Then again since Doppio and Diavolo are split personalities, they're essentially the same person.
That and the whole fate thing are a bit too vague to properly give his character a boost on a first read.
Every other villain had a clearer motive and/or theme than he had.
Oh and he had the shit luck of his stand stealing all the spotlights from him simply by working.
