For a comedy harem, 4 to 5 allows for the best variety of archetypes, designs and characterization without cast bloating.
Most comedy harems see an appalling drop in quality after they go beyond 5, because the cast becomes too large to maintain a stable dynamic and balance screentime. new characters end up either competing with old ones for screentime, invading an otherwise stable scenario and destabilizing it, or being completely redundant and all-but replacing a member of the old cast.
Most harems become trapped in this paradigm where their only good, interesting or funny arcs are character introductions, and so they're forced to continuously introduce new characters just to have something to write about, until the cast becomes bloated and they have to find an excuse to sideline old characters (which alienates their fans) in favour of palette-swap replacements and create the mid-story cast replacement paradigm that marks the death knell of a harem series.

Generally the paradigm is 4 main girls and a 5th that is either an occasionally-appearing side-character, or primarily outside of the main love situation and just there to shake things up and keep things exciting.

Since 6 is right in the middle of the sequence i would say this is the perfect number, just enough to have a flavor for everyone but not enough to make some of the girls get overshadow'd, also even number so no girl gets to stay in the front in the promo images and make it obvious who will "win"
For the main harem, 4-5 is probably the best. But I think you can get away with 9-10 girls if you had them separate into groups.
