In order to be a gun servant, you have to be a true skilled guy without question.

Mark of Cain would be a worthless power for a Grand Servant. Grand Servants are supposed to fight against the ultimate threats to the planet. If they lose, that's it. Final game over. Who cares if the thing that's threatening the planet dies after the fact? By that point, there's nothing left to protect.
It's also questionable if such a concept would even work against literally everything. Gilgamesh was unaffected by all the evils of the world and it's possible that things like TYPEs might be too alien for human curses to work against.

A Grand Assassin's would probably need a powerset that's a little more generalized than the average Assassin's. Something that doesn't lose any effectiveness against non-human targets, that isn't reliant on things like human physiology or weaknesses. Being strong against humans isn't what's important here. If that's all that was necessary, then the planet can just sic Primate Murder on it. A Grand Assassin needs to be someone who can kill "anything", not just humans.
