The joke is that Iihiko was a stand-in for the reader and thus could not be affected by any physical stuff that happens in the manga and could beat any opponent by just taking a page and ripping it in half. Only narrative bullshit could affect him, since narrative is the only way a work of fiction can affect (emotionally) the reader.

When I said skills I specifically meant the "skills" (abnormalities/minuses) in Medaka Box.
I doubt knives would have any effect on him because of how durable he is, Medaka had to break the sound barrier to deal any damage, so even outside of his immunity to skills he is incredibly strong and durable.
In the end it ends up being his own attack that destroys him due to his immense strength and "irreversible destruction" ability.

Niemani's ability is a style instead of a skill, which means Iihiko isn't invulnerable to it.
Something about styles working through words and words being able to reach Iihiko since they have conversations with him.
The reason why Nienami's style specifically is such a big deal is because it's completely broken with it giving the least possible outcome of an event.
Since Iihiko is almost indestructible, this means the least possible outcome of someone hitting him would be him instantly dying, therefore a single hit from Zenkichi after being powered up with Nienami's style would be fatal.

Kumagawa has a couple powers during the series but the main one is ALL FICTION, which can make anything (from concepts to events to things) into nothing, completely removing them from existence.
If you're asking about his inability to win, that's because the Minuses have really bad luck, they can win a fight but only in a way that will be unsatisfactory and won't feel like them actually winning, like winning because of the enemy's mistakes.

kumagawa will not 'always' lose, you see him demolish characters left right and center. his position as a minus is that he will always lose to main characters, while defeating side characters with ease. He is the final boss character, no matter how strong he appears he is trapped in the role of a strong enemy whose eventual destiny is to lose to the power of friendship when it comes to the final battle with the MC and their friends.

Even when fighting side-characters and winning with ease he always finds a way to spin it into him losing.
He "loses" against Aka, the cadets, Ezumachi and Sukinasaki even though he beats them.
Munakata is also a side character, but Kumagawa lost to him.
It's not that he's unable to win, it's just that he has extremely poor luck and lacks the confidence to feel he ever won properly.
