There would be one campaign but it would drastically scale in difficulty depending on who you play as.
IE:

Instead of the super bar being filled on hit it is filled with time, it can be used every 10 seconds or when Saitama has less than 10hp left ( out of 1000 or so ). It will oneshot any opponent if they dont block and only Boros can barely survive it when blocking. Dodging it is possible though but you still take heavy damage, unless you manage to teleport behind saitama or something.

Humans legitimately require rest between exercise sessions for the dead muscle to be recovered and rebuilt. Otherwise the muscle won't recover enough to grow, or will get fatigued enough to tear, or other bad things will happen.
Saitama didn't rest. He repeatedly destroyed his body, which is incredibly stupid but in this case gave him super powers.

So in the last episode Saitama destroys the meteor and said thats the best he thinks he can do. Does that mean Saitama does not believe that after destroying the meteor he does did believe he had the ability to destroy all the rocks before they fell?
Though thinking about it it would of been much better if he did that Boros serious punch on the ground. The air pressure alone would of been enough to destroy the meteor into dust.

Easy mode is make it a regular musou/fighting game/beat them all like all the Marver/DC games where Superman/Wolverin somehow can get beaten up by regular punks
Normal mode is make it beat-them-all where you only rarely play as Saitama
Hard mode that will never happen:
Make the gameplay centered on collateral damages.
You'd play as Saitama in a sandbox city, kinda like Saints Row 4.
Everything is destructible and random events happen all the time.
You need to rise in popularity and rank.
Saving civilians raises your popularity, taking down monsters raises your rank.
However injuring civilians and destroying buildings as collateral will drop your popularity and mitigate your rank gains, making fighting big and strong opponents really tricky.
Other heroes can also join the fights and steal the credit.
Sometimes it would be impossible to fight a huge monsters without doing a mess and thus you'd only lose point by taking them out, but if you just avoid the encounter to keep your rankings a district would be destroyed before another hero stops it, and that area would basically be devoid of any content until it's rebuilt.
So on the long run if you don't bite the bullet, the sandbox will be mostly ruins.
Anyway that's an interesting gameplay I would really like to see for an OPM game
