I take thousands of screenshots of all my anime and then organize them by giving them descriptive names, so that only the finest screenscots for use as reactions reach your eyeballs from my end. Pic related.

I don't know about like, but when I see an interesting grimace in an anime, I have the following criteria before judging it worth working on:
- The head shouldn't cut off at the top of the video frame. In fact, the more the part I want is shaped by the frame's outlines, the less I am interested.
- No color bleeds. When the color from the background (or the lighting) notably creeps into the part that I want. When you remove the background it looks like shit.
- Sharp outlines. Otherwise the transparency won't come out good. I can improvise with feathering and blur effects, but that will only go so far.
- not too much smug. I aim to have only one smug image in every 5 reaction images that I produce. I don't have a good reason for this rule.

of course smug anime girls and shitty stream crops have their appeal as well for other reasons than aesthetics

generally something that has a pretty immediate effect, like you don't have to look at it twice to get it, or open it or what have you.
While I do like a reaction image that's funny or memorable, I think they're best used to compliment or convey a message

That would usually go into "surprise that you care" or "why do you still care" (with positive or negative reaction respectively) or a simple look of boredom. I usually don't go with boredom because if I'm actually bored, I'll stop responding.
Then there's the option of somebody just enjoying themself with the implication being that the topic isn't interesting enough to distract them from whatever they're doing. Pic related.
