In a generic Japanese high school, an entire freshman class magically changes sex one night. All the boys become girls and all the girls become boys. They retain memories of themselves and their classmates original sex, but no one else (friends, parents, teachers) does. Now they need to adapt to their new lives and teach each other how to live as their new sex.
There wouldn't be one main character but instead every episode would be about a different character or characters and how they're dealing with it, their relationships with people inside and outside of their class, etc.
Different episodes could have different themes. Some would be comedy, some romance, some drama/angst, something a mix of everything.

Angsty episode: There's a yuri couple in the class, a tomboy and a feminine chick that is afraid of men. They both become men and while tomboy doesn't mind it too much, feminine chick keeps freaking out and there's angst as she doesn't want to have gay sex with her girlfriend since she's afraid of penises. Eventually they break up and tomboy starts dating the former class ume turned girl.
Romance/comedy episode: A bunch of former girls, now guys, decide to take revenge on the class pervert, now a girl, by trying to turn tables on him and molest him, just like he always tried to molest them in the past. Unfortunately for them, their plan backfires as he realizes what they're doing and plays along, much to their surprise and disgust.
And to make matters more complex, one of the girls in class was infatuated with him for a long time and confesses to him while all this is happening in the background. He doesn't take her seriously and plays along, thinking she's one of the girls trying to mess with him, and they end up on a date. She's ecstatic about it, and during the date he slowly starts to realize she wasn't messing around and genuinely wants a relationship, even with the two of them being the wrong sex.
Another: A girl in class had a very close relationship with her older sister. They would trade clothes, talk about boys, etc. Now that she's a guy (and from her sister's PoV she was always a guy - altered memory), she's trying to figure out how to maintain her formerly close relationship with her sister. The episode is told from her older sister's PoV, who is freaking out because her otouto, who was always distant from her (altered memory), is suddenly acting all friendly and uncomfortably girly and she thinks he's trying to come out of the closet to her.
Then another episode about the class delinquent (guy) turned class idol trying to deal with his former gang (all in other classes) no longer respecting him but instead hitting on him.

A comedy about an actual normal high school guy while all his classmates are like main characters from other anime. So we'd have some classmates trying to revive their baseball team and make nationals, some in a shoujo love triangle, some hunting super powered ghosts, some secretly magical girls and so on and so forth while MC is stuck in the middle of all of this and only relevant because he sits in 'that' chair.
